RK-Spectro Technical Note 63: Single wavelength kinetic of cytochrome c in fluorescence mode

Introduction

The objective of this technical note is to realize folding kinetics of cytochrome c at different temperatures: 25, 20, 15 and 10°C. Folding kinetics were initiate d by a 6-fold dilution of unfolded cytochrome c in 4.5M of Guanidine hydrochloride (pH 5.89 before mixing and final pH of 6.76 after folding).
The scenario documented presupposes a good understanding of the MOS-200 instrument. Bio-Kine version 4.52 was used to control the syringes, edit the sequences, follow the acquisition and to analyze results (fit calculation).
